方法
1. Western blotting
Western blotting法检测P53靶向抑制剂-EGFR对人结肠癌细胞株（HT-29,colo205）中，p53/p27信号通路下游关键因子p27的蛋白表达情况，并将人结肠癌细胞株区分为p27高表达的细胞株和低表达的细胞株。
Methodology
1. Western blotting
The protein expression of the p53/p27 signal pathway downstream key factor p27 in the treatment of human colon carcinoma cell strain（HT-29,colo205）by P53 targeted inhibitor EGFR was examined by the Western blotting method, and the human colon carcinoma cell strains were separated into p27 high expression and low expression cell strains.
2. MTT和RT-PCR
MTT法测定EGFR对结肠癌细胞转染前后抑制率的变化，RT-PCR法测定p27沉默前后，mRNA的表达量的变化,根据实验结果进行统计分析。
2. MTT and RT-PCR
While the detection of the changes in inhibition rate of EGFR before and after colon carcinoma cell transfection was conducted by the MTT method, the RT-PCR method was used to detect mRNA expression level changes before and after P27 silencing, and then a statistic analysis was conducted based on the experiment results.
